✏️ Recently, the emergence of genome-editing technologies including ZFN, TALEN, and CRISPR-Cas9 systems, has sparked a revolution in cervical cancer-targeted therapy.

✏️Delivery is one of the biggest challenges in genome-editing therapeutic development.

✏️Cas9 may be delivered to cells in DNA, mRNA, or protein format, and each mode has unique advantages, disadvantages, and delivery requirements.

✏️The combination of nanocarrier systems, anti-cancer drugs, imaging agents, and CRISPR/Cas9 systems can offer a unique opportunity for the development of future cervical cancer treatment strategy.

🔆DNA Methylation of CD70 Promoter in Juvenile Systemic Lupus Erythematosus

✅Epigenetic alterations in pathogenesis of systemic lupus erythematosus (SLE) have gained more attention in adults recently.

✅We assessed the methylation of CD70 promoter, a costimulatory molecule on T cells, in juvenile SLE (JSLE).

✅Twenty-five patients with JSLE and 24 healthy controls were compared. JSLE patients had lower unmethylated CpG islands compared to the control group (mean ± SD; 0.78 ± 0.42 vs 10503.80 ± 39796.95). However, the difference was not significant (P-value; 0.22).

✅Despite hypomethylation of CD70 gene promoter in CD4+ T-cells from adult patients with SLE, no statistically significant differences observed in patients with JSLE compared with healthy controls. This may suggest a mechanism different in JSLE patients than in adults.

Pro-tumorigenic functions of macrophages at the primary, invasive and metastatic tumor site

🌀 The tumor microenvironment (TME) not only facilitates cancer progression from the early formation to distant metastasis, but also it difers itself from time to time alongside the tumor evolution.

🌀Tumor-associated macrophages (TAMs), whether as pre-existing tissue-resident macrophages or recruited monocytes, are an inseparable part of this microenvironment.

🌀As their parents are broadly classifed into a dichotomic, simplistic M1 and M2 subtypes, TAMs also exert paradoxical and diverse phenotypes as they are settled in diferent regions of TME and receive diferent microenvironmental signals.

Development and clinical application of bispecific antibody in the treatment of colorectal cancer

New paper by USERN interest groups

🌀Treatment of colorectal cancer as one of the most commonly diagnosed and a frequent cause of cancer-related deaths is of great challenges in health-related issues.

🌀Immunotherapy is the fourth pillar of cancer treatment which provides more novel therapeutic options with expanding investigational potentials.

🌀One of the modalities in immunotherapy is the use of bispecific antibodies. Despite demonstrating many promising roles, it still needs more advanced studies to identify the actual pros and cons.

🌀In this review, the application of bispecific antibody in the treatment of colorectal cancer has been explained, based on preclinical and clinical studies.

🌀Bispecific antibody is in its early stages in colorectal cancer treatment.

Novel treatments using PD1 inhibitors for advanced and metastatic cutaneous squamous cell carcinoma

🌀Cutaneous squamous cell carcinoma (SCC) accounts for the second most commonly occurring non melanoma skin cancer.

🌀Metastatic and locally advanced cutaneous SCC have poor response to surgery or radiotherapy; however, systemic therapies can be an alternative option for patients that are not candidates for local therapies.

🌀The combination of PD1 inhibitors with other therapies such as radiation may increase the response in advanced cutaneous squamous cell carcinoma.

🌀Administration of PD1 inhibitors have shown higher response rates in tumors with high TMB compared to those with lower tumor mutation burden; However, other variables must be considered.
