BCGitis as the primary manifestation of chronic granulomatous disease

🌀Patients with primary immunodeficiency disease (PID) are not only vulnerable to mycobacterial disease, but are also more likely to develop adverse events following BCG vaccination. These events can range from regional disease (BCGitis) to disseminated disease (BCGosis). Chronic granulomatous disease (CGD), which is characterized by impaired leukocyte phagocytic function, is one of the many inherited PIDs that increase the body’s susceptibility to recurrent bacterial and fungal infections. Here, we report a 6-year-old boy with no significant past medical history who presented with progressive lymphadenopathy six years after BCG vaccination. He was later diagnosed with CGD on further evaluation.

Chimeric Antigen Receptor T-cell Therapy for Melanoma

🔶🔷Highlights:

🔹Melanoma is the deadliest of skin cancers.
🔹There are several therapeutic options for melanoma which have not reached the maximum efficacy.
🔹CAR T cell therapy is a type of ACT which showed efficacy in animal models for tumor cells resistant to other approaches.
🔹CAR T cell has limited efficacy for solid tumors.
🔹Choosing an accessible and stable tumor antigen and potent co-stimulatory molecules is necessary to improve the outcome.
🔹Several antigens including GD2, CD20, CD70, gp100, NY-ESO-1, B7-H3, and VEGFR-2 have been tested for targeting by CAR T cells for melanoma treatment in clinical studies.
🔹The results of currently ongoing clinical trials to assess CAR T cell will determine its efficacy for melanoma.

Recent progress in immunotherapy of breast cancer targeting the human epidermal growth factor receptor 2 (HER2)

🌀Breast cancer is responsible for most of the cancer-induced deaths in women around the world. The current review will discuss different approaches of targeting HER2, an epidermal growth factor overexpressed in 30% of breast cancer cases.

🌀As well as the introduction of HER2 and breast cancer subtypes, we discussed various aspects of HER2-targeting immunotherapy including monoclonal antibodies, Antibody-drug conjugates (ADCs), Chimeric Antigen Receptor (CAR) T-cells and vaccines.

🌀Despite several ways of controlling breast cancer, the need to investigate new drugs and approaches seems to be much significant as this cancer still has a heavy burden on people’s health and survival.

COVID-19 in patients with cancer: Risks and precautions

New Paper on COVID-19 by USERN Interest Group

🌀It has been reported that those who suffer from cancer (especially lung cancer and hematological malignancies) are at higher risk of serious complications and death from COVID-19.

🌀Some cancer treatments such as CAR T cell therapy can produce a cytokine storm, which is also a hallmark of severe COVID-19. Therefore, patients receiving CAR T cells are at higher risk if they become infected with COVID-19, and could be treated with anti-cytokine approaches.

♨️Current and Future Perspectives of PD-1/PDL-1 Blockade in Cancer Immunotherapy

🌀Cancer immunotherapy especially immune checkpoint inhibitors have been of particular interest and have gained approval by the FDA for treatment of cancers.

🌀Blockade of programmed cell death protein-1 (PD-1) has demonstrated promising antitumor effects for many different solid and hematologic malignancies. However, despite promising results, a favorable response is observed only in a fraction of patients, and there is still lack of a single therapy modality with curative ability.

🌀In this paper, we review the current and future perspectives of PD-1/L1 blockade in cancer immunotherapy, with a particular focus on predictive biomarkers of response to therapy, adverse events associated with PD-1/L1/2 inhibitors and the potential strategies for improving the efficacy of immunotherapy with PD-1/L1 checkpoint inhibitors.
